Product Overview | This ELISA Kit employs the Sandwich-ELISA method. The micro ELISA plate provided in this Kit has been pre-coated with an antibody specific to POT1. To conduct the assay, standards or samples are introduced into the respective micro ELISA plate wells and mixed with the specific antibody. Then a biotinylated detection antibody specific for POT1 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each microplate well successively and incubated. Unbound components are then thoroughly washed away. Following this, a substrate solution is introduced to each well. Only those wells that contain POT1, biotinylated detection antibody, and Avidin-HRP conjugate will appear blue in color. The enzymatic reaction with the substrate is halted by adding a sulfuric acid solution, causing the color to change to yellow. The optical density (OD) is quantitatively measured using a spectrophotometer at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of POT1. You can determine the POT1 in the samples by comparing the OD of the samples to the standard curve. |

Notes | Occasionally, trace amounts of reagent may remain in the closure of the product vial during shipment and storage of the POT1 ELISA kit. It is recommended that any liquid that may be present in the lid of the container be centrifuged to the bottom using a bench-top centrifuge prior to experimentation. |

Is a component of the double-stranded telomeric DNA- binding TRF1 complex which is involved in the regulation of telomere length by cis-inhibition of telomerase. Also acts as a single-stranded telomeric DNA-binding protein and thus may act as a downstream effector of the TRF1 complex and may transduce information about telomere maintenance and/or length to the telomere terminus. Component of the shelterin complex (telosome) that is involved in the regulation of telomere length and protection. Shelterin associates with arrays of double-stranded TTAGGG repeats added by telomerase and protects chromosome ends; without its protective activity, telomeres are no longer hidden from the DNA damage surveillance and chromosome ends are inappropriately processed by DNA repair pathways.
