Product Overview | This immunoassay Kit enables the quantitative determination of target antigen concentrations in vitro, using serum, plasma, tissue homogenates, cell culture supernates, or other biological fluids as specimens. The microtiter plate provided with this Kit comes pre-coated with an antibody that specifically recognizes the target antigen. Following this, standards or samples are introduced into the respective microtiter plate wells, along with a biotin-conjugated antibody specific to the target antigen. Subsequently, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and allowed to incubate. The next step involves the addition of a TMB substrate solution to each well. Only those wells containing the target antigen, biotin-conjugated antibody, and enzyme-conjugated avidin will display a color change. To conclude the enzymatic reaction, a sulfuric acid solution is added, and the color shift is quantitatively measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of the target antigen in the samples is then determined by comparing the optical density (O.D.) of the samples to the standard curve. |

Notes | Occasionally, trace amounts of reagent may remain in the closure of the product vial during shipment and storage of the TERT ELISA kit. It is recommended that any liquid that may be present in the lid of the container be centrifuged to the bottom using a bench-top centrifuge prior to experimentation. |

UniProt Comments | Telomerase is a ribonucleoprotein enzyme essential for the replication of chromosome termini in most eukaryotes. Active in progenitor and cancer cells. Inactive, or very low activity, in normal somatic cells. Catalytic component of the teleromerase holoenzyme complex whose main activity is the elongation of telomeres by acting as a reverse transcriptase that adds simple sequence repeats to chromosome ends by copying a template sequence within the RNA component of the enzyme. Catalyzes the RNA-dependent extension of 3'-chromosomal termini with the 6-nucleotide telomeric repeat unit, 5'-TTAGGG-3'. The catalytic cycle involves primer binding, primer extension and release of product once the template boundary has been reached or nascent product translocation followed by further extension.
