TK102 Human Protection of Telomeres Protein 1 (POT1) ELISA Kit

This ELISA Kit employs the Sandwich-ELISA method. The micro ELISA plate provided in this Kit has been pre-coated with an antibody specific to POT1. To conduct the assay, standards or samples are introduced into the respective micro ELISA plate wells and mixed with the specific antibody. Then a biotinylated detection antibody specific for POT1 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each microplate well successively and incubated. Unbound components are then thoroughly washed away. Following this, a substrate solution is introduced to each well. Only those wells that contain POT1, biotinylated detection antibody, and Avidin-HRP conjugate will appear blue in color. The enzymatic reaction with the substrate is halted by adding a sulfuric acid solution, causing the color to change to yellow. The optical density (OD) is quantitatively measured using a spectrophotometer at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of POT1. You can determine the POT1 in the samples by comparing the OD of the samples to the standard curve.

The test principle applied in this Kit is Sandwich enzyme immunoassay. The microtiter plate provided in this Kit has been pre-coated with an antibody specific to Protection Of Telomeres 1 Homolog (POT1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Protection Of Telomeres 1 Homolog (POT1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Protection Of Telomeres 1 Homolog (POT1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Protection Of Telomeres 1 Homolog (POT1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This Kit utilizes the Enzyme-Linked Immunosorbent Assay (ELISA) method. The plate has been pre-coated with human TRF1 antibody. TRF1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human TRF1 Antibody is added and binds to TRF1 in the sample. Following this, Streptavidin-HRP is introduced and binds to the Biotinylated TRF1 antibody. During a washing step, any unbound Streptavidin-HRP is removed. A substrate solution is then added, and color development occurs in proportion to the quantity of human TRF1. The reaction is halted by adding an acidic stop solution, and the absorbance is measured at 450 nm.
