This ELISA Kit employs the Sandwich-ELISA method. The micro ELISA plate provided in this Kit has been pre-coated with an antibody specific to POT1. Then a biotinylated detection antibody specific for POT1 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each microplate well successively and incubated. Unbound components are then thoroughly washed away. Following this, a substrate solution is introduced to each well. Only those wells that contain POT1, biotinylated detection antibody, and Avidin-HRP conjugate will appear blue in color. The enzymatic reaction with the substrate is halted by adding a sulfuric acid solution, causing the color to change to yellow. The optical density (OD) is quantitatively measured using a spectrophotometer at a wavelength of 450 nm +/- 2 nm. The OD value is proportional to the concentration of POT1. You can determine the POT1 in the samples by comparing the OD of the samples to the standard curve.
